Artesunat-e (INN) is part of theartemisinin group of drugs that treat malaria. It is asemi-synthetic derivative of artemisinin that is water-soluble andmay therefore be given by injection. It is sometimes abbreviatedAS.
Synthesis
Artesunat-e is prepared fromdihydroartemisinin (DHA) by reacting it with succinic acidanhydride in basic medium. Pyridine as base/solvent, sodiumbicarbonate in chloroform and catalyst DMAP (N,N-dimethylaminopyridine) and triethylamine in 1, 2-dichloroethanehave been used, with yields of up to 100%. A large scale processinvolves treatment of DHA in dichloromethane with a mixture ofpyridine, a catalytic amount of DMAP and succinic anhydride. Thedichloromethane mixture is stirred for 6–9 h to get artesunat-e inquantitative yield. The product is further re-crystallized fromdichloromethane. alpha-Artesunat-e is exclusively formed (m.p135–137°C).
